چکیده

T HE anti-thyroid drugs, thiouracil and 6 n-propylthiouracil have been reported to cause leukopenia and granulocytopenia in man and experimental animals. ‘ Some evidence has been found to support the view that concurrent administration of liver preparations and folic acid may have beneficial effect on the toxicity of thiourea and thiopyrimidines, including the blood dyscrasias.’#{176} 2 In view of these reports and since Petering, Delor and Murray’3 had shown that concurrent administration of sufficiently large doses of folic acid (pteroylglutamic acid-PGA) retarded the onset of sulfonamide-induced blood dyscrasias in the rat and lessened their severity, it was of considerable interest to carry out similar studies in the rat on the thiopyrimidine-induced blood dyscrasias. It should be emphasized that this work differs from that of other investigators in that the antithyroid drugs and the folic acid preparations were administered together during the entire course of the experiments.

Treatment with folic acid ameliorated the histological and immunohistochemical alterations caused by propylthiouracil –induced hypothyroid rat aorta

Thyroid status is an important determinant of cardiovascular function. We studied the changes in the aorta of hypothyroid rat at the post-pubertal stage, in addition to the ameliorating role of folic acid. Fifty male albino rats were equally divided into five groups; the first and second groups were the control and folic acid groups, respectively while the third group was the hypothyroid group ...

Does Propylthiouracil Increase the Gentamicin-Induced Nephrotoxicity In Rat?

Objective(s): The aim of this study was to evaluate the effect of subacute administration of propylthiouracil (PTU) on gentamicin (GM)-induced nephrotoxicity in male rats.   Materials and Methods: Male Wistar rats were divided into 4 experimental groups as follow: (1) Control group: isotonic saline (1 ml/kg, IP. for 18 d), (2) GM group: 100 mg/kg, IP for 8 d, (3) PTU group: PTU...

Reaction between Thiouracil derivatives and Chloroasetic acid in gas and soluble phases:A theoretical study

Thiouracilis a historically relevant anti-thyroid preparation. Because of its structure you can find it in various chemical reactions differently. In this study, the reaction of Thiouracil with Chloroacetic acid and the formation of their additive products has been investigated. This reaction is aconcerted process, and it has not been determined yet by exhaustive mechanisms. From the potential ...

Limited response to propylthiouracil in the tumor-bearing rat.

The accidental observation that the thyroid gland of the tumor-bearing rat was not enlarged after the oral administration of thiouracil was made while investigating the possible role of the thyrotrophic hormone (TSH) as a fat-mobilizing hormone (14). Further experiments were carried out to ensure that the effect was not limited to a particular strain of rat or type of tumor, or due to the actio...

The action of pteroyl glutamic acid and natural sources of folic acid on blood dyscrasias induced by sulfonamide drugs.

By H. G. PETERING, PH.D., R. A. DELOR, B.S., AND H. C. MURRAY, PH.D. F OLIC acid or vitamin B9 deficiency has been induced in the rat by feeding a purified diet containing succinyl sulfathiazole or sulfaguanidine)’2’3’4’5’6 This deficiency is corrected in either case by feeding free or conjugated pteroyl glutamic acid while the syndrome produced by sulfaguanidine is also corrected by paraaminob...
